DNA is negatively charged and therefore the dyes chosen need to be negative as well in order for the dyes to move in the same direction and be as close as a to a genuine DNA test. Based on the results, Brilliant Blue, Orange G, Cresol Red, Xylene Cyanol, Phenol Red, Bromocresol green, Bromophenol blue and Bromocresol Purple are negatively charged and thus selected. Toluidine blue and M-cresol purple were not chosen since they are positive dyes and moves in the opposite direction.
1.5% agarose gel is firmer and easier handle thus making it more user friendly which is important if the kit was to be sold to the public. Moreover the Dyes tend to stay longer in the1.5% agarose compared to 1% agarose gel., this is due to the higher concentration resulting in smaller pores and traps the dye for a longer period of time.
